A novel roseobacter phage possesses features of podoviruses, siphoviruses, prophages and gene transfer agents

Abstract: Bacteria in the Roseobacter lineage have been studied extensively due to their significant biogeochemical roles in the marine ecosystem. However, our knowledge on bacteriophage which infects the Roseobacter clade is still very limited. Here, we report a new bacteriophage, phage DSS3Φ8, which infects marine roseobacter Ruegeria pomeroyi DSS-3. DSS3Φ8 is a lytic siphovirus. Genomic analysis showed that DSS3Φ8 is most closely related to a group of siphoviruses, CbK-like phages, which infect freshwater bacterium C… Show more

“…Roseophage SIO1 and DSS3Ф8 also possess the phoH gene [13,26]. Phage-encoded phoH genes have previously been described as apparent parts of a multi-gene family with divergent functions and have played a part in phospholipid metabolism, RNA modification, and fatty acid beta-oxidation [54,61,62].…”

“…All phages with both genes have relatively large dsDNA genomes (from Ruegeria phage DSS3P8 of 146,135 bp to Cronobacter sakazakii phage GAP32 of 358,663 bp). Nearly all of the phages with the two-gene system are Myoviridae phages, with the exception of the Siphoviridae phage DSS3P8 and the large Siphoviridae Caulobacter crescentus phiCbK and phiCbK-like phages (13,14). Clustal Omega alignments and neighbor-joining trees ( Fig.…”

“…However, despite the ecological significance of the Roseobacter clade (Luo & Moran, ), only a small number of roseophages have been isolated and described (Liang et al., ). The few characterized roseophages are assigned to either the Podoviridae or Siphoviridae families, comprising both lytic and temperate phages (Ankrah, Budinoff, Wilson, Wilhelm, & Buchan, ; Liang et al., ; Zhan, Huang, Voget, Simon, & Chen, ) and are considered a key driving force for the genomic diversification of the Roseobacter clade (Zhan et al., ).…”
