A Novel Photosensitizer 31,131-phenylhydrazine -Mppa (BPHM) and Its in Vitro Photodynamic Therapy against HeLa Cells

Abstract: Photodynamic therapy (PDT) has attracted widespread attention due to its potential in the treatment of various cancers. Porphyrinic pyropheophorbide-a (PPa) has been shown to be a potent photosensitizer in PDT experiments. In this paper, a C-3 1 ,13 1 bisphenylhydrazone modified methyl pyropheophorbide-a (BPHM) was designed and synthesized with the consideration that phenylhydrazone structure may extend absorption wavelength of methyl pyro-pheophorbide-a (Mppa), and make the photosensitizer potential in deep t… Show more

“…MPPa belongs to the second generation of photosensitizers and is a chlorophyll derivative (30). To date, MPPa-PdT has been used clinically for the treatment of a variety of tumors such as osteosarcoma, lung and cervical cancer (17,19,31). The present study aimed to explore the functional mechanism of MPPa-PdT in osteosarcoma.…”

“…As described in our previous report, 41 the mechanism of photochemical processes (Type-I and Type-II) in PDT was investigated by quenching the corresponding ROS generated from the photodynamic reaction using quenching agents. For the Type-I mechanism, a series of ROS such as superoxide, hydrogen peroxide and hydroxyl radical are directly produced during the photochemical process after irradiation (the hydroxyl radical also could be generated from an interaction between superoxide and hydrogen peroxide by the Fenton/ Haber-Weiss reactions, 42 which were not explored in detail here as it was beyond the scope of this article).…”

“…Examining Table II we may see that 16 N S is more significant than Q 16 . Therefore, and as a first approximation, we shall not consider Q 16 . Atom 20 is a carbon atom of the side chain of ring C (Figure 2).…”

“…16 shows that a high inhibitory activity is related with a positive value for Q 16 , fact that seems to be contradictory with the interaction with an electron deficient center. Examining Table II we may see that 16 N S is more significant than Q 16 . Therefore, and as a first approximation, we shall not consider Q 16 .…”
