A novel necroptosis-related gene index for predicting prognosis and a cold tumor immune microenvironment in stomach adenocarcinoma

Abstract: BackgroundGastric cancer (GC) represents a major global clinical problem with very limited therapeutic options and poor prognosis. Necroptosis, a recently discovered inflammatory form of cell death, has been implicated in carcinogenesis and inducing necroptosis has also been considered as a therapeutic strategy.ObjectiveWe aim to evaluate the role of this pathway in gastric cancer development, prognosis and immune aspects of its tumor microenvironment. Methods and resultsIn this study, we evaluated the gene exp… Show more

“…Among all the signatures, StepCox [backward] + Enet [alpha = 0.7] showed the highest average C-index value (0.726). This signature, named in this study as IRS, is composed of 24 genes: TNFAIP2 39 , 40 , SLC37A2 , RGS1 41 , ZNF101 , TM6SF1 , CRHBP , AKAP5 42 , CRYBB1 , S100Z 43 , ACSM5 , NTAN1 44 , IL5RA , ABCG1 , CAMK4 , MCEMP1 45 47 , SLC2A3 48 50 , RENBP , BASP1 51 , 52 , KYNU 53 , 54 , CTLA4 55 , FCGR2B 56 , ENTPD8 , DDO , FCN1 57 . All of these genes have been mentioned in previous studies to affect the prognosis of patients with GC.…”

“…APOD is a potential biomarker of hypoxia and is involved in immune responses ( 20 ). APOD is included in a novel necrosis-related gene model for predicting the prognosis of gastric adenocarcinoma and is closely associated with the immune microenvironment of cold tumors ( 21 ). In glioma, POSTN may regulate resistance to anti-VEGF-A therapy by upregulating the expression of TGFβ1 and HIF1α ( 22 ).…”
