A novel model of distal colon cancer in athymic mice

Abstract: . Conception, design and critical revision. ABSTRACT PURPOSE:The present a novel adenocarcinoma model in athymic mice. METHODS:Seven athymic mice were used. Colon diversion and distal fistula were made. Adenocarcinoma cells were inoculated in the submucosa of fistula. Tumor growth was monitored daily. Scintigraphy with 99mTc-MIBI was performed to identify the tumor. RESULTS:The model of distal colon cancer is feasible. Tumor detection was possible by both, macroscopically and molecular imaging. All resections d… Show more

“…To explore the pathogenic pathway of specific bacteria, implantation of human colon cells in immunosuppressive nude mice would be an ideal method (11,12). Previous studies have described an open surgical method of implanting human colon cells into mice (12,(31)(32)(33); nevertheless, this method involves challenging techniques and operative complications (12,31). Transanal submucosal injection is nonoperative and is less invasive than the open surgical method (12,13,16,17).…”

“…The procedures were performed in a laminar flow with rigorous asepsis and antisepsis techniques [ 45 ]. On day 0, to perform the cell inoculation, mice were anesthetized with xylazine hydrochloride 2% plus ketamine, diluted 1:2 (0.3 mL/20 g), and administered via intramuscular injection.…”

“…The culture medium was changed every 24 h. Cell viability was evaluated using trypan blue staining. HT-29 cells (4 × 10 6 ) were suspended in 40 μL of normal saline and directly seeded into the subcutaneous tissue of the back left flank of athymic mice through a percutaneous puncture (27-gauge hypodermic needle) using a 1 mL syringe [ 68 , 72 , 73 ].…”

“…After xeno-transplantation, tumor growth was checked daily using metal calipers. The formula “Volume = L × S × S/2”, where “S” is the minor diameter measured and “L” is the largest diameter measured, was used to determine the CC volume [ 68 , 72 , 74 ].…”
