A novel method for the quantification of fatty infiltration in skeletal muscle

Abstract: BackgroundFatty infiltration of the skeletal muscle is a common but poorly understood feature of many myopathies. It is best described in human muscle, where non-invasive imaging techniques and representative histology have been optimized to view and quantify infiltrating fat. However, human studies are limited in their ability to identify cellular and molecular mechanisms regulating fatty infiltration, a likely prerequisite to developing targeted interventions. As mechanistic investigations move to small anim… Show more

“…An Electron‐Multiplying Charge‐Coupled Device cooled camera (Hamamatsu; Quorum Technologies, Guelph, 80 ON, Canada) and Volocity 6.3 software (PerkinElmer, Waltham, MA, USA) were used to capture and analyse all images. Visualization and quantification were performed as previously described . Z‐stacked images captured tissue sections using a 20×/0.85 oil lens and were assembled together and plane merged to create a composite image that enabled the visualization of a whole and clear tissue cross section.…”

“…As previously described (39), whole diaphragms were decellularized in 1% SDS for 5 days, fixed in 4% PFA, and then incubated in Oil red O (ORO) dissolved to 0.3% by weight in 60% isopropanol. After overnight incubation in 1% SDS, they were affixed to wax plates or dissection petri dishes for analysis.…”

“…15 Sections were stained with hematoxylin and eosin (H&E) and Picrosirius Red according to standard protocols described previously. 16 Overlapping images were taken of each H&E-stained section and digitally registered to reconstruct the full TA cross-section. Fibers with centralized nuclei were identified and manually marked on reconstructions.…”
