A novel label-free fluorescence assay for one-step sensitive detection of Hg2+ in environmental drinking water samples

Abstract: A novel label-free fluorescence assay for detection of Hg2+ was developed based on the Hg2+-binding single-stranded DNA (ssDNA) and SYBR Green I (SG I). Differences from other assays, the designed rich-thymine (T) ssDNA probe without fluorescent labelling can be rapidly formed a T-Hg2+-T complex and folded into a stable hairpin structure in the presence of Hg2+ in environmental drinking water samples by facilitating fluorescence increase through intercalating with SG I in one-step. In the assay, the fluorescen… Show more

“…Different techniques have been used for Hg 2+ detection, such as the fluorescence detection technique, 137 Atomic Absorption Spectroscopy (AAS), 138 Ti 3 C 2 T x -FET, 139 etc. Among them detection with the MXene-FET took only 10 s and it detects 5 ppm mercury ion rapidly, which is efficient.…”

“…Since such reaction has high sensitivity and selectivity, it has also been used to develop the reaction-based fluorescent probe of Hg 2+ ions. 130–134 For example, Li et al 135 had designed a novel label-free fluorescence assay based on T-rich DNA, which can form a T–Hg 2+ –T stable hairpin structure in the presence of Hg 2+ ions. The resulting T–Hg 2+ –T complex can facilitate fluorescence enhancement via intercalating with SYBR Green I (SG I), which acted as a fluorescent dye that had been widely used in analytic fields (Fig.…”

“…Microchannels can be easily built into cloth using wax screen printing. Lab on cloths are simple and cost effective while combining the properties of conventional lab on a chip detection systems [127]. Liu et al used flow chemiluminescence for chromium detection in water using a lab on cloth device.…”
