A novelabi5allele reveals the importance of the conserved Ala in the C3 domain for regulation of downstream genes and salt tolerance during germination inArabidopsis

Abstract: Abscisic acid (ABA) signal transduction during Arabidopsis seed development and germination requires a Group A bZIP transcription factor encoded by ABA INSENSITIVE5 (ABI5). In addition to the basic leucine zipper DNA binding domain, Group A bZIPs are characterized by three N-terminal conserved regions (C1, C2 and C3) and one C-terminal conserved region (C4). These conserved regions are considered to play roles in ABI5 functions; however, except for the phosphorylation site, the importance of the highly conserv… Show more

“…Since ABI3 is known to interact with ABI5 and other group A bZIP proteins (Hobo et al, 1999;Nakamura et al, 2001;BrocardGifford et al, 2003;Finkelstein et al, 2005;Marella et al, 2006;Park et al, 2011;Tezuka et al, 2013), our results together with the previous findings suggest that ABI3, ABI5, and DELLAs interact with each other on the SOM promoter to activate SOM mRNA expression at high temperature.…”

“…Thus, seed germination is strongly inhibited at high temperature. bZIPs, including ABI5 and PIL5 (Hobo et al, 1999;Nakamura et al, 2001;Brocard-Gifford et al, 2003;Finkelstein et al, 2005;Park et al, 2011;Tezuka et al, 2013), their enrichment could be facilitated by the formation of a protein complex between ABI3 and other transcription factors on the SOM promoter. Such proteinprotein interactions could strengthen an otherwise weak targeting of transcription factors to a promoter.…”

“…We transiently expressed the constructs in Arabidopsis mesophyll protoplasts and monitored transcriptional activity via LUCIFERASE (LUC) expression driven by the EM6 promoter (Fig. 8C), a well-defined ABI5-responsive promoter (Carles et al, 2002;Tezuka et al, 2013). As was observed in our yeast one-hybrid assays, the expression of wild-type ABI5 induced more than a 3-fold increase in LUC activity relative to the vector control (Fig.…”

“…Previous studies showed that ABI3 interacts with ABI5 and regulates an overlapping set of physiological events as a regulatory complex (Nakamura et al ., ; Tezuka et al ., ). Thus, we further investigated the relationship between the ABI5–BES1 and ABI3–ABI5 interactions.…”
