A Novel Ecdysone Receptor Mediates Steroid-Regulated Developmental Events during the Mid-Third Instar of Drosophila

Abstract: The larval salivary gland of Drosophila melanogaster synthesizes and secretes glue glycoproteins that cement developing animals to a solid surface during metamorphosis. The steroid hormone 20-hydroxyecdysone (20E) is an essential signaling molecule that modulates most of the physiological functions of the larval gland. At the end of larval development, it is known that 20E—signaling through a nuclear receptor heterodimer consisting of EcR and USP—induces the early and late puffing cascade of the polytene chrom… Show more

“…45 These proposals would be in line with another study showing that silencing USP in the salivary gland does not block glue protein synthesis but it requires EcR. 46 Yet, the binding of ecdysone to a receptor, other than the heterodimer EcR/USP, remained to be proven.…”

“…Dominant-negative (EcR-DN) and RNAi (EcR-RNAi) UAS transgenes targeting EcR were expressed in larval salivary glands using the salivary gland Gal4 driver (sgGal4) (Costantino et al, 2008), and semi-quantitative RT-PCR was performed to determine levels of pri-let-7-C in -18 hours, -8 hours, -4 hours and 0 hour APF RNA samples. Expression of EcR-DN or EcR-RNAi greatly diminished pri-let-7-C levels in third larval instar salivary glands (Fig.…”
