A novel dsRNA element isolated from the Aspergillus foetidus mycovirus complex

Abstract: Aspergillus foetidus virus (AfV) contains at least two icosahedral particle types named AfV-fast (-F) and AfV-slow (-S), based on relative electrophoretic mobility. AfV-F is a quadripartite double-stranded RNA (dsRNA) virus, and AfV-S contains AfV-S1, which is a member of the genus Victorivirus in the family Totiviridae, and AfV-S2, which may be a satellite RNA or satellite virus and is described here. Analysis of the complete AfV-S2 nucleotide sequence reveals it to be significantly similar to an unclassified… Show more

“…A similar organization is reported for RnMV-2, also referred to as yado-kari virus 1 [58], whereas AfsV-2 has a smaller genome and the conserved RdRp domain is positioned in the center of the protein [59]. Both, RnMV-2 and AfsV-2 are described to be unique viruses which hijack the capsid protein of an unrelated dsRNA virus [58,59]. Whether this unusual behavior also holds true for RsMV-1 needs further investigation.…”

“…S4B). Mycovirus RdRPs are known to be related to the RdRPs of viruses in the picorna-like superfamily of eukaryotic, positive-strand RNA viruses (26), and numerous examples can be found in the literature including Ustilaginoidea virens nonsegmented virus 1 (27), a number of viruses in the Partitiviridae family (28), and Aspergillus foetidus virus S2, which is closely related to several members of the Caliciviridae family and Comovirinae subfamily (29). Our phylogenetic analysis results described here (Fig.…”

“…7). PaFlV1 instead forms a wellsupported clade with Aspergillus foetidus slow virus 2 that stems from a ssRNA tree, therefore showing that this two viruses are indeed phylogenetically ssRNA viruses, and the classification of Aspergillus foetidus slow virus 2 as a dsRNA virus element (Kozlakidis et al, 2013) should, in our opinion, be reviewed. Supplementary Fig.…”
