A novel culture method that sustains ERα signaling in human breast cancer tissue microstructures

Abstract: Background Estrogen receptor α (ERα) signaling is a defining and driving event in most breast cancers; ERα is detected in malignant epithelial cells of 75% of all breast cancers (classified as ER-positive breast cancer) and, in these cases, ERα targeting is the main therapeutic strategy. “…Institutional Review Board (IRB) approval (IRB STU00202331) was obtained from Northwestern University prior to consenting patients and collecting samples. To prepare breast microstructures the protocol of Brisken's group was used with some minor modifications (33,34). The unaffected contralateral breast tissues of postmenopausal women who underwent bilateral mastectomy due to incident unilateral breast cancer were obtained while fresh and diced to approximately 5 mm pieces before digesting at 37°C with 2% collagenase I in F12-K nutrient mix (Kaighn's) medium, overnight.…”
