A Novel cis-Acting Element in Promoters of Plant B-Type Cyclin Genes Activates M Phase-Specific Transcription

Abstract: Plant B-type cyclin genes are expressed late in the G2 and M phases of the cell cycle. Previously, we showed that the promoter of a Catharanthus roseus B-type cyclin, CYM, could direct M phase-specific transcription of a beta-glucuronidase reporter gene in synchronously dividing BY2 tobacco cells. In this study, we determined the regulatory elements contained within the CYM promoter by using a luciferase reporter gene. Mutational analysis showed that a 9-bp element is essential for M phase-specific promoter ac… Show more

“…Whether functional properties of NtE2F resemble the E2F-1, 2 and 3 or E2F-4 and 5 subfamilies, along with whether there are related genes in plants, remains to be determined. Although Ito et al [15] reported that a M-speci¢c activator is a common cis-acting element that controls the M phasespeci¢c expression of cell cycle-related genes, the control of G1 and S phase-speci¢c expression is poorly understood in plants. In mammalian cells, the E2F/DP family of transcription factors is thought to play important roles in gene regulation during the S phase [7].…”

“…In our study, the cyclin B promoter was then variously modified, by deletion or point mutation, and assayed for its ability to direct phase-dependent expression of the luciferase reporter gene in BY2 cells (Ito et al 1998). This experiment identified an important cis element, called MSA element, which is repeated four times in the C. roseus cyclin B promoter.…”

“…These 3RMyb proteins were found to bind in vitro to the DNA sequences specified by the cis element with the core sequence of AACGG. This cis element is known as M phase-specific activator (MSA) element, and was first identified in tobacco (Ito et al 1998). cDNAs for two other 3RMyb proteins, NtmybC1 and NtmybC2, were isolated recently by PCR with the primers designed for amino acid sequences in the Myb domain that are highly conserved among 3RMyb proteins.…”

“…1) were required to confer G 2 ͞M-specific expression in the context of the Ϫ205 promoter. Previous work had shown that this motif is required for G 2 ͞M phase expression in Catharanthus roseus cyclin CYM, when assayed in BY2 cells (17,34). The AACGG core MSA motif was mutated to ACTAG or AGCTG.…”
