A Nonproducer T Lymphoblastoid Cell Line from Marek's Disease Transplantable Tumor (JMV)

Abstract: A continuous lymphoblastoid cell line was established from a JMV tumor transplant related to Marek's disease (MD). It is designated RPL1 (JMV) lymphoblastoid cell line. This cell line contains DNA sequences complementary to MD virus DNA and has an antigen similar to MD-tumor-associated surface antigen (MATSA). However, it lacks any MD virus (MDV) rescuable in vivo or in vitro. The cell line has surface antigens typical of chicken thymus cells (T cells) and histocompatability antigens different from those of th… Show more

“…Ten chicken cell lines established from chicken lymphoma were used in this study, MD cell lines: MSB1 [1], MTB1 [9], JP1, JP2 [31], HP1, HP2 [20] and RP1 [18]; AL cell lines: 1104B1 [6] and CU10 [3] an RE cell line: Ku7 [22]. All cell lines were grown at 40°C in RPMI1640 medium supplemented with 10% fetal bovine serum, penicillin G (200 U/ml), streptomycin (200 mg/ml) and 2 × 10 5 M 2-mercaptoethanol.…”

“…DT95 and DT40 are c-myc-transformed chicken B-cell lines (Baba et al, 1985). MSB-1 and RP-1 are T-cell lines derived from lymphomas induced by Marek's disease virus (Akiyama and Kato, 1974;Nazerian et al, 1977). BM2 is a macrophage-like cell line derived from avian myeloblastosis virus (AMV) transformed yolk sac cells (Moscovici et al, 1982).…”

“…This highly malignant tumour kills chicks in 5 to 8 days, much less time than is required for induction of tumours by inoculation of the virus or virus-producer cell lines such as MSB 1. The RP 1 and BP 1 cell lines are derived from this JMV tumour, and have a T-ceU marker and MATSA, but lack any MDV rescuable in vivo or in vitro (Nazerian et al, 1977;Sekiya et al, 1977). On the other hand, the MSB1 and HP1 line cells, which are derived from MD lymphomas, also have a T-cell marker and MATSA, but produce MDV.…”

“…However, the exact nature of MATSA is not clearly understood. Dissimilarity or partial qualitative differences of MATSA in different MD cell lines have been noted (Witter et al, 1975;Sharma et al, 1977;Nazerian et al, 1977;Sugimoto et al, 1979;Rennie & Powell, 1979). Recently we demonstrated the presence of Hanganutziu and Deicher (H-D)-like heterophile antibody in chicken anti-MATSA sera (Ikuta et aL, 1980).…”
