In this study, we focused on studying the changes in urine metabolites in hyperlipidemic rats using ultra‐performance liquid chromatography coupled with quadrupole time‐of‐fight mass spectrometry (UPLC–Q‐TOF/MS) and metabolomics, as well as the effect of Citri Reticulatae Chachiensis Pericarpium (CRCP) on hyperlipidemia. These urine samples were examined by UPLC–Q‐TOF/MS to obtain MS data. The MS data were analyzed by principal component analysis and partial least squares‐discriminant analysis to identify the differential metabolites. CRCP reduced the body weight and levels of triglycerides, total cholesterol and low‐density lipoprotein cholesterol and abnormally decreased high‐density lipoprotein cholesterol in hyperlipidemic rats, which were significantly raised by a high‐fat diet. Twenty‐seven potential biomarkers were identified within the complex sample matrix of urine. Fourteen biomarkers increased in the hyperlipidemia rats compared with normal rats. Meanwhile, 13 biomarkers decreased. CRCP reversed abnormal changes in biomarkers, including 5‐l‐glutamyl‐taurine, 5‐aminopentanoic acid, cis‐4‐octenedioic acid and 2‐octenedioic acid. These biomarkers show that hyperlipidemia is related to the metabolic pathways of taurine and hypotaurine metabolism, fatty acid biosynthesis, and arginine and proline metabolism. CRCP mainly prevents hyperlipidemia by intervening in these metabolic pathways.

Aerial parts of Astragalus membranaceus (APAM), which are derived from the overground part of Astragalus membranaceus consist of stems and leaves. The aim of this study was to provide a scientific basis for the comprehensive utilization of APAM. Nuclear magnetic resonance (NMR) was used to compare the chemical compositions of APAM and Astragali Radix (AR). The pharmacological effects of APAR and AR in cyclophosphamide (Cy)-induced mice were studied using 1 H NMR-based metabolomics. The results showed that APAM and AR were different from each other in primary and secondary metabolites, and they could recover the amounts of white blood cell, monocytes, lymphocyte and neutrophils content, IL-2 and IL-6; there was no significant difference in APAM and AR in terms of immune functions. The mechanisms of APAM and AR on leucopenia were probably related to energy metabolism, amino acids metabolism, and gut microbiota-related metabolism. ARTICLE HISTORY

Metabolic disturbances have been associated with many human diseases, including cancer, diabetes, and cardiovascular disease. Metabolomics, a rapidly growing member of the –omics family, investigates cellular metabolism by quantifying metabolites on a large scale and provides a link between metabolic pathways and the upstream genome that governs them. With the advances in analytical technologies, metabolomics is becoming a powerful tool for identifying diagnostic biomarkers of diseases, elucidating the pathological mechanisms, discovering novel drug targets, predicting drug responses, interpreting the mechanisms of drug action, as well as enabling precision treatment of patients. In this review, we highlight the recent advances of technologies and methodologies in metabolomics and their applications to the field of clinical pharmacology. Recent publications from 2013 to 2018 are covered in the review, and current challenges and potential future directions in the field are also discussed.
