A non-steroidal anti-inflammatory drug (ketoprofen) does not delay β-TCP bone graft healing

Abstract: Beta-tricalcium phosphate (beta-TCP) is a suitable biomaterial in oral and maxillofacial surgery since it can induce a rapid proliferation of woven bone. Granules, prepared by the polyurethane foam method, were implanted in critical size defects performed in the femoral condyles of New Zealand rabbits. Animals were studied after 8 and 28 days. Ketoprofen (a non-steroidal anti-inflammatory drug (NSAID)) was given for 8 and 28 days to evaluate its effects on the healing of the graft. Before euthanasia, the rabbi… Show more

“…Numerous studies have reported that the biocompatible and osteoconductive b-TCP scaffolds could support the attachment, proliferation, and differentiation of osteogenic cells (such as osteoblasts and bMSCs). 27,[33][34][35][36] In agreement with these literatures, the microscopy analysis of the bMSCs/b-TCP constructs in this study also showed that the cells could adhere to the surface of the scaffold, spread, and proliferate. As in regular culture on tissue culture plastic, the bMSCs cultured on b-TCP scaffold displayed the expected osteogenic activity when cultured in the osteogenic medium.…”

“…An in vivo study evidenced active osteoblasts on newly formed trabeculae and active osteoclasts 2 weeks after implantation β‐TCP particles in the rat 13. Similarly, our group evidenced the centripetal development of new bone trabeculae by microcomputed tomography as early as 8 days after implantation in the femoral condyle of the rabbit 26. After a 3‐month implantation period, the extensive formation of bone trabeculae formed onto and around β‐TCP particles is reported in a number of studies 27–30.…”

“…Analysing these studies further, we could not identify any association among the class or potency of the studied NSAIDs to inhibit the COX-1 or COX-2 enzyme, the dose, or the timing. In terms of timing, although some authors suggest that short-term administration after fracture could be safe, others contradict this finding suggesting that NSAID administration is safe only if it is initiated a few weeks after fracture [78, 85, 98, 101, 105, 106, 108, 126]. A link, however, can exist between the size of the animals.…”
