A non-curved chicken lysozyme 5' matrix attachment site is 3' followed by a strongly curved DNA sequence

Abstract: Matrix attachment regions (MARs) partition the genome into functional and structural loop-domains. Here, we determined the relative matrix affinity of cloned fragments of the chicken lysozyme 5' MAR. We show that this region contains a non-curved high-affinity binding site, which is 3' followed by a strongly curved DNA sequence that exhibits weak matrix binding. DNA curvature is not a physical property required for strong matrix binding. Possible biological functions of this sequence arrangement, particularly … Show more

“…This construct was made by use of standard molecular biology techniques 22 and was grown in Escherichia coli strain STBL2 (Life Technologies). We coinjected a 2-fold molar excess of the Ϸ3-kb chicken lysozyme matrix attachment region 23 to improve specificity of transgene expression. 24 Transgenic mice were prepared in the University Transgenic Mouse Facility, with FVB/N mice used as donors and recipients.…”

“…The direction of the bend, in turn, may determine whether YY1 acts as an activator or repressor (30). Both parameters, DNA-bending potential (3,16,31) and strand separation potential (5) are not only functionally related but are also properties that have been associ- FIG. 10.…”

“…Currently it is believed that DNA structure (perhaps a narrow minor groove, a tendency to form bent DNA, a tendency for single-strandedness and a tendency to form looped structures) is responsible for the matrix binding activity of a region (36)(37)(38)(49)(50)(51)(52). The data indicating that MARs may be enriched in inverted repeats (reviewed in 38) are of particular interest.…”
