A newly discovered gene, tfuA, involved in the production of the ribosomally synthesized peptide antibiotic trifolitoxin

Abstract: Trifolitoxin (TFX) is a gene-encoded, posttranslationally modified peptide antibiotic. Previously, we have shown that tfxABCDEFG from Rhizobium leguminosarum bv. trifolii T24 is sufficient to confer TFX production and resistance to nonproducing strains within a distinct taxonomic group of the ␣-proteobacteria (E. W. Triplett, B. T. Breil, and G. A. Splitter, Appl. Environ. Microbiol. 60:4163-4166, 1994). Here we describe strain Tn5-2, a Tn5 mutant of T24 defective in the production of TFX, whose insertion maps… Show more

“…Few rhizobia are known to produce certain metabolites such as antibiotics (7,21), siderophores (3) and HCN (23) which play a significant role in rhizosphere affecting the growth and activity of the other microbes and plant health as well. Based on results, M. loti MP6 produces hydroxamate type of siderophore.…”

“…The PatD C terminus (PatD2) is similar to YcaO-like conserved domains of unknown function but also to SagD from Streptococcus iniae, which may serve as a hydrolase (43). PatD2 shows similarity to TtufA, a protein involved in the synthesis of the ribosomally derived trifolitoxin (44). The entire PatD peptide sequence is similar to only a handful of proteins, including the T. erythraeum homolog, a protein annotated as AknN (a hydrolase) from Streptomyces galilaeus, and Orf12, a predicted protein of unknown function from the granaticin biosynthetic pathway (45).…”

“…TvaB, TvaM, and TvaN are categorized as regulatory proteins based on sequence similarities. TvaI is also assignable as a regulator, because a homologous protein, TfuA, can regulate the production of trifolitoxin, a RiPP family antibiotic (19).…”
