A New Vero Cell Rabies Vaccine: Results of a Comparative Trial with Human Diploid Cell Rabies Vaccine in Children

Abstract: We evaluated the immunogenicity and safety of a chromatographically purified rabies vaccine (CPRV) compared with human diploid cell rabies vaccine (HDCV) after pre-exposure immunizations (both primary and booster). Intramuscular doses of either 0.5 mL of CPRV or 1.0 mL of HDCV were given to 400 schoolchildren on days 0, 7, 28, and 365 (booster). Adequate titers of antibody (> or = 0.15 IU/mL, as defined by the Centers for Disease Control and Prevention) were observed in serum samples from all children 14 days … Show more

“…BPL-inactivated virus vaccines against yellow fever [63], SARS-CoV [64], influenza [65] and rabies [66] were able to induce the production of neutralizing antibodies. However, BPL-inactivated influenza showed reduced hemagglutinin and neuraminidase activity [62].…”

“…Previously published studies with larger sample sizes have consistently shown a lower frequency of local and systemic reactions with CPRV in relation to HDCV. 14,17 This trial showed comparable frequencies of local reactions and slightly higher frequencies for systemic reactions occurring within 3 days of CPRV administration. The occurrence of mild myalgia after dose 1 was higher in the CPRV group (56%) than in the HDCV group (40%).…”

“…Previously published studies 14,17 have tended to show lower GMTs with CPRV than with HDCV. However, this is thought to be clinically irrelevant, as both groups were well above the minimally acceptable seroconversion titer of Ն 0.5 IU/mL.Despite this, the GMTs reported after dose 3 in this study were similar (Table 4).…”

“…It is known to be a well-established platform in the field of vaccine development (Delrue et al, 2012). Various vaccines, based on this platform, have been developed which include vaccines for polio, influenza and rabies with a safe record (Delrue et al, 2012;Murdin et al, 1996;Sabchareon et al, 1999;Vellozzi et al, 2009). For SARS-CoV-2, there have been 10 vaccines in clinical phase and 11 vaccines in pre-clinical phase as listed by WHO, on 9 th march 2021, (World Health Organization, 2021a).…”
