A new solution for an old problem: the regiochemical distribution of the acyl chains in galactolipids can be established by electrospray ionization tandem mass spectrometry

Abstract: Electrospray ionization-quadrupole ion trap mass spectrometry (ESI-QITMS), either in positive- or in negative-ion mode, has been used to establish the chemical structures (chain length, degree of unsaturation, positional distribution) of the fatty acids attached to the primary (sn-1) and secondary (sn-2) hydroxyl groups of the glycerol moiety of natural monogalactosyl- (MGDG) and digalactosyldiacylglycerols (DGDG), isolated from the freshwater dinoflagellate Glenodinium sanguineum and from a marine diatom belo… Show more

“…The fatty acyl groups were identified based on the fragments deriving from neutral loss (NL) of individual fatty acyl groups. Fragments corresponding to the NL of sn-2 fatty acyl groups are known to yield lower abundances than those from the NL of sn-1/3 fatty acyl groups (Guella et al, 2003); thus, the abundances of tandem mass spectrometry product fragments were used for regiochemical determination.…”

“…Under this chromatographic setup, lipid molecular species were separated primarily according to the hydrophobicity of their acyl chains. Details are reported in Guella et al (2003). Hydrophilic Interaction LIquid Chromatography (HILIC) was also used to establish the membrane lipid class composition based on the different polarity of lipids head groups (aneSi & Guella 2015).…”

“…The positions of the acyl chains (sn-1 or sn-2) were determined by their relative percent compositions according to a variation of the procedure (i.e. similar conditions on a different instrument) established by Guella et al (2003).…”
