Abstract. Aryantini D, Astuti P, Yuniarti N, Wahyuono S. 2023. Bioassay-guided isolation of the antioxidant constituent from Kaempferia rotunda L. Biodiversitas 24: 3641-3648. Kaempferia rotunda L. contains various phytochemical compounds with various biological activities and has been widely utilized in traditional medicine. This research focused on exploring bioactive compounds in the ethanol extract of Kaempferia rotunda using radical scavenging (1,1-Diphenyl-2-picrylhydrazyl) bioassay-guided isolation. Initially, the powdered material of K. rotunda was macerated with 70% ethanol and filtered, and the filtrate was evaporated in vacuo to make concentrated ethanol extract. The concentrated ethanol extract was then triturated gradually with increasing polarity of solvents (n-hexane, ethyl acetate, ethanol) to give n-hexane (HSF), ethyl acetate (EASF), ethanol (ESF), and residue (ISF) fractions. Each fraction was tested by DPPH bioassay with quercetin as the positive control to determine the active fraction. The active fraction (ESF) was further fractionated by Vacuum Liquid Chromatography (VLC) using a mobile gradient phase starting from ethyl acetate 100%, acetone 100%, and methanol 100% tomake3 sub-fractions (F1, F2, F3),respectively. The DPPH radical scavenging bioassay showed that F1 was the most active, containing bioactive compound detected by TLC visualized by DPPH. Based on spectroscopic and literature data comparison, this compound was isolated, purified, and identified ascrotepoxide. Crotepoxide displayed IC50of 38.91±0.59 (ABTS), 47.45±0.60 (DPPH),and 26.74±1.23 (FRAP) µg/mL.

In this study, we aimed at evaluating in vitro and in vivo anti-inflammatory activity of various extracts of the rhizomes of Globba pendula Roxb. Three extracts ( n-hexane, ethyl acetate, and water) were screened for their inhibitory effect on NO production by lipopolysaccharide-stimulated RAW 264.7 macrophages. The ethyl acetate extract of G. pendula rhizomes (EGP) showed a potential effect with an IC50 value of 32.45 µg/mL. For in vivo study, the ethyl acetate extract was further investigated for its anti-inflammatory effect using collagen antibody-induced arthritic mice (CAIA). The level of arthritis in experimental mice significantly reduced ( P < .05) after treatment with EGP at a dose of 500 mg/kg body weight (b.w.). This study also revealed that EGP is orally non-toxic. Ethyl p-methoxy cinamate was identified as the main constituent of EGP, which may result in its anti-inflammatory effect.

Globba schomburgkii Hook.f. rhizomes were extracted with dichloromethane and fractionated using column chromatography to obtain 10 fractions (FA-FJ). The most active fractions, FE and FG, were further fractionated to give seven and thirteen sub-fractions, respectively. The fractions and sub-fractions were tested for antibacterial activity using 96-well plate microdilution method. The antibacterial activity of several sub-fractions (such as FE.B, FE.C, FE.D, FG.D, FG.G, and FG.H) were higher than that of the crude extract. Chemical compositions of all fractions and sub-fractions were analyzed by gas chromatography hyphenated with mass spectrometry (GC-MS), revealing 167 compounds identified according to match between the experimental MS and retention index data and that from the NIST17 library. The major volatile compounds were identified as ?-bicyclohomofarnesal and (E)-15,16-dinorlabda-8(17),11-dien-13-one (P2). Some of the compounds such as (E)-labda-8(17),12-diene-15,16-dial, methyl isocostate, zederone, alantolactone, and 8?,11?elemadiol have been reported to be biologically active. Lastly, two pure compounds isolated by column chromatography were characterized by 1H, 13C, COSY, HSQC and HMBC NMR techniques as petasol (P1) and P2.
