This study aimed at evaluating phytoplankton as a monitoring tool for water quality assessment in an urban shallow eutrophic reservoir considering temporal and vertical scales. Garças Reservoir is located in the Parque Estadual das Fontes do Ipiranga Biological Reserve (23°38 0 08 00 S and 23°40 0 18 00 S; 46°36 0 48 00 W and 46°38 0 00 00 W) that lies in the southeastern part of the Municipality of São Paulo, southeast Brazil. Samplings were carried out monthly during 8 consecutive years (1997)(1998)(1999)(2000)(2001)(2002)(2003)(2004) following the water column vertical profile (5 depths: subsurface, 1, 2, 3 m and *20 cm from the bottom). Abiotic variables analyzed were: water temperature, electric conductivity, DO, pH, total alkalinity, free CO 2 , dissolved inorganic carbon, N series, P series and SiO 4 H 4 . Biological variables studied were: total density, total biomass and chlorophyll a, which were integrated arithmetically. Abstract-The aquatic environment often contains different groups of contaminants, but their combined toxicity on microalgae has seldom been reported. The present study compared the toxic effects of combined mixed polycyclic aromatic hydrocarbons (PAHs) and heavy metals on growth and antioxidant responses of free and immobilized microalga, Selenastrum capricornutum. Five PAHsphenanthrene, fluorene, fluoranthene, pyrene, and benzo[a]pyrene-and four heavy metals at different concentrations-0.05 to 0.1 mg Cd 2þ ml À1 , 0.05 to 1 mg Cu 2þ ml À1 , 0.05 to 1 mg Zn 2þ ml À1 , and 0.5 to 2.5 mg Ni 2þ ml À1 -were examined. Results showed that the chlorophyll a content of free and immobilized S. capricornutum was not affected by PAHs but was significantly inhibited by heavy metals. Conversely, the antioxidant parameters, including the content of reduced glutathione (GSH) and the activities of superoxide dismutase and peroxidase, were significantly induced by both PAHs and metals. For the combined toxic effects of PAHs and heavy metals, cell growth and antioxidant responses varied with exposure time and contaminants and differed between free and immobilized cells. The effects of cocontaminants on the GSH content in free cells were mainly synergistic but changed to antagonistic in immobilized cells. The toxic effects of cocontamination on free cells were also more obvious than those on immobilized cells. These findings suggest that immobilization offers some protection to microalgal cells against toxic contaminants causing differences in the interaction and responses to combined toxicants between free and immobilized cells. Abstract:Chlorophyll is the green pigment in plants that helps to absorb sunlight and converts it into energy. It is believed that it is beneficial for the human body. Therefore in this study, the chlorophyll amount in three brown algae species (Sirophysalis trinodis, Polycladia myrica and Colpomenia sinuosa) of the Persian Gulf by using three solvents and applying two formulas was compared. Acetone, diethyl ether and methanol were used as solvents. Results showed that the chlorophyll amount in different species dependent on solvent type and cell wall structure of algae. The highest chlorophyll a and x+c contents were observed in S. trinodis species. Also, the highest content of chlorophyll c 1 +c 2 was showed in C. sinuosa by the extraction with diethyl ether solvent. In both formulas was no significant difference between the same solvents in the amount of chlorophyll a. In general, the aim of this study due to the diversity and dispersion of algae and also their potential use in the food industry, cosmetics and pharmaceutical goods was to choose a suitable solvent for better extraction of chlorophyll as an active ingredient. Hence, in this study, the acetone was a better solvent.
