A new isoquinolone alkaloid from an endophytic fungus R22 of Nerium indicum

Abstract: A new isoquinolone alkaloid named 5-hydroxy-8-methoxy-4-phenylisoquinolin-1(2H)-one (3), together with two known quinolinone alkaloids 3-O-methylviridicatin (1) and viridicatol (2) were isolated from the fermentation of an endophytic fungus Penicillium sp. R22 in Nerium indicum. Their structures were elucidated by NMR, IR and MS data, and were also confirmed by comparing with the reported data in the literature. Meanwhile, the antibacterial and antifungal activities of all compounds were tested, and the result… Show more

“…Vancomycin treatment was associated with enrichment of pathways for isoquinoline compounds in both CDI and non-CDI stool samples; the enrichment of this pathway regardless of CDI status suggests that this observation represents an antibiotic-induced community function. These heterocyclic aromatic compounds are used in the production of antimicrobial agents ( 31 ), including antifungal agents ( 32 ).…”

“…R22 associated with Nerium indicum collected from Qinling Mountain, Shaanxi Province, China was the source of 5-hydroxy-8-methoxy -4-phenylisoquinolin-1(2H)-one ( 145 ) a new isoquinolone alkaloid along with 3- O -methylviridicatin ( 146 ) and viridicatol ( 147 ) ( Figure 7 ) two known quinolinone alkaloids. Compound 145 exhibited good antifungal activity against Alternaria brassicae , A. alternata and Valsa mali with MIC value of 31.2 µg/mL, compound 146 against A. brassicae , B. cinerea and Valsa male with MIC value of 31.2 µg/mL, compound 147 against A. brassicae , A. alternata and B. cinerea with MIC value of 31.2 µg/mL [ 79 ].…”

“…#12 in >10-fold higher quantity than the ΔlaeA mutant, and pyripyropene O was putatively identified and produced in >2-fold higher quantity. Cyclopenol is an alkaloid that is described as a mycotoxin in the benzodiazepine class 70 and serves as an biosynthetic intermediate for viridicatol 71 , which has been described as having antibacterial activity against Staphylococcus aureus 72 . Small amounts of viridicatol were indeed found in extracts with high quantities of cyclopenol.…”
