Protein extracted from Douchi was hydrolyzed by alcalase based on a singlefactor experiment and response surface methodology was employed to optimize hydrolysis conditions. The optimum conditions were determined to be: 63℃ extraction temperature, 1.4% quantities of enzyme and substrate, and 1.7 h extraction time. The optimal protein hydrolysate was subsequently subjected to ultrafiltration, and the fraction of molecular weight 10-50 kDa was identified as the most potent contributor to antioxidant activity. Amino acid analysis indicated that acidic amino acids were the most abundant in all fractions of the protein hydrolysate. Infrared spectroscopy showed there were no significant differences in secondary structure between fractions.

Marine environment is a huge reservoir of various important resources for new bioactive molecule discovery from marine organisms. The aim of this investigation is to evaluate the antiproliferative potential of crude proteins extracted from different organs of Donax variabilis (gills, adductor muscles, mantle and whole tissue) on human Lung (A549), oral (KB3-1), breast (MCF-7) and colon (HT-29) cancer cell lines. The crude proteins extracted from four different organs of marine clam were biochemically and qualitatively analysed using Native and SDS PAGE. These extracts were tested against normal (vero) cell line to determine the toxicity of crude proteins. Among the four organs, proteins from mantle potentially inhibited growth of lung, oral and breast cancer cell lines in a dose dependent manner and inhibition of growth in colon cancer cell line was time dependent when examined by MTT assay. Significant inhibition of cell viability on A549 cells observed at (IC 50) 100 µg/ml, KB 3-1, MCF-7 cells at (IC 50) 250 µg/ml concentration after 24 h of incubation and HT-29 at (IC 50) 500 µg/ml concentration after 48 h of incubation. The crude (mantle) protein treated cancer cell lines showed modifications in cell morphological features. Further investigation on nuclear morphology was done by propidium iodide staining under fluorescence microscope and DNA ladder assay showed fragmented pattern of DNA that confirms induction of apoptosis. On observation, it can be concluded that crude extract (mantle) from D. variabilis highly effective against human cancer in-vitro. INTRODUCTION: Over past few decades cancer is one of the most lethal diseases in human being and it has become a worldwide health issue. Cancer is the generic term for a large group of disease characterized by the growth of abnormal cells and they can invade other organs beyond their usual boundaries.

Cancer is a big public health problem in many parts of the world. A novel anti-tumor protein (J2-C2) was previously isolated from Arca inflata and it was reported that this protein had antiproliferative effect on some human tumor cell lines such as A549, HepG2 and SPC-A-1. In this study, firstly, J2-C2 was produced by recombinant techniques in the Escherichia coli strain BL21 (DE3) pLysE and this protein was purified by Ni-NTA agarose affinity chromatography. Expressed recombinant J2-C2 was analyzed with SDS-PAGE. 75.5 mg ml-1 of J2-C2 was achieved from a 600 mL culture. Then using HT-29, MCF7 and PC3 cancer cell lines, we showed the effect of recombinant of J2-C2 on cell proliferation, migration and apoptosis in a cell specific manner. Cell viability was measured using MTT assay. Additionally, real-time-qPCR was applied to analyze the transcript levels of apoptosis related genes such as Bcl-2, Bax and p53. The 2-ΔΔCt method was performed to determine the relative changes in gene transcription. Moreover, scratch wound healing assay was performed to evaluate the effect of J2-C2 on cancer cell migration. Consequently, we found that recombinant J2-C2 did not have a significant effect on cell viabilities of MCF7, PC3 and HT29 in concentration-dependent manner. Furthermore, our results showed that recombinant J2-C2 declined HT29, MCF7 cell migration. However, we did not observe the same results for PC3 cancer cell line.
