A new hybrid resin for stepwise screening of peptide libraries combined with single bead Edman sequencing

Abstract: A library system was developed for the discovery of bioactive peptides. Library synthesis and peptide sequencing was performed on a solid support while the screening for bioactivity was done with peptides in solution. The peptides were synthesized by split and mix, one-bead-one-peptide library synthesis, using a Tentagel S-NH2 solid support with a loading of approximately 100 pmol/bead. The major part of the peptide was connected to the support by a single acid-labile linker and a minor part of the peptide was… Show more

“…16 The library was screened using the three-step converging screening system as described. 15 In the initial screening round, two peptide pools induced proliferation (SI of 10 and 20, respectively). The peptide-bearing beads from the pool with the highest induction of proliferation were used in the second round of screening.…”

“…Design, synthesis and use of a dedicated synthetic peptide library Hybrid TentaGelH-AM resin (particle size, 90 mm; loading, 100 pmol/bead, 16 pmol acid stabile attached, 84 pmol acid labile attached) 15 was used to synthesize a random onebeadFone-peptide 14-mer peptide library containing a DPw4 binding motif. 16 The hybrid resin allows for a convergent library screening using the acid cleavable part of the peptide material attached to the resin, combined with efficient peptide identification by Edman sequencing or MS-MS mass spectrometryy, using the noncleavable part of the peptide material attached to the resin.…”

“…The beads are microporous and consist of a polystyrene core onto which polyethyleneglycole (PEG) is grafted to a final content of 50 to 70% (w/w). 26 The narrow pore size of TentaGel resins typically prevents fluorescently labeled target proteins, usually larger than 15 kDa, from penetrating into the bead interior within the typical incubation and screening time. This confines the majority of the relevant signal to the bead periphery, i.e.…”

“…One such resin is TentaGel, a graft copolymer of a crosslinked polystyrene matrix and polyethylene glycol [21]. TentaGel beads have been used extensively in biological assays with combinatorial peptide libraries and show very low non-specific protein adsorption [9, 11]. In this study, we demonstrate a process that combines phage display discovery of mimetope peptides with solid phase immunoassay methods for the quantification of mAb levels in biologic samples.…”
