A New Class of Tungsten-Containing Oxidoreductase in Caldicellulosiruptor, a Genus of Plant Biomass-Degrading Thermophilic Bacteria

Abstract: bCaldicellulosiruptor bescii grows optimally at 78°C and is able to decompose high concentrations of lignocellulosic plant biomass without the need for thermochemical pretreatment. C. bescii ferments both C 5 and C 6 sugars primarily to hydrogen gas, lactate, acetate, and CO 2 and is of particular interest for metabolic engineering applications given the recent availability of a genetic system. Developing optimal strains for technological use requires a detailed understanding of primary metabolism, particularl… Show more

“…A 10-base sequence of the promoter was repeated on each side of the IS element. Expression of the aor gene from P. furiosus (Pfaor) did not appear to be negatively affected, as 127 bp of the 3= end of the promoter were still intact, and the expression level of Pfaor was higher than that of the native slp gene (12).…”

“…The first occurred when a strain, MACB1002, was constructed to heterologously express the gene (aor) encoding the aldehyde oxidoreductase of Pyrococcus furiosus, driven by the slp promoter on a replicating shuttle vector (12). The replicating plasmid was isolated from the recombinant strain, but the plasmid size differed from that of the original transformation plasmid by about 1.5 kb.…”

“…Cultures of strain UFO1 (50 ml) were grown to late log phase (20 h) in modified R2 broth without (in triplicate experiments) and with (in duplicate experiments) 50 M uranium added to the growth medium. RNA was isolated from the cultures and reverse transcription-PCR (RT-PCR) was performed as previously described (32). Primers for RT-PCR are listed in Table 1.…”

“…For example, it has been used in developing a replicating shuttle vector for C. bescii and other Caldicellulosiruptor species (9), and this vector has been used for homologous overexpression of the genes celA, encoding a bifunctional glycoside hydrolase (10), and xor, encoding a tungsten-dependent oxidoreductase (11). Numerous gene deletions in addition to cbeI have also been constructed, including the ldh gene encoding lactate dehydrogenase (12), a gene cluster encoding pectin-degrading enzymes (13), the celA gene (14), and a gene cluster encoding [Ni-Fe] hydrogenase maturation enzymes (15).…”
