Endometrial cancer is one of the most commonly diagnosed cancers in women. The search for factors that contribute to the development of cancer cells in reproductive organs should involve the detection of xenoestrogens, in particular zearalenone (ZEA) and its metabolites. Xenoestrogens are endocrine disruptors-ZEA and its metabolites are structurally similar to estrogens (macrocyclic lactone ring) and show high affinity for estrogen receptors. This study proposes a new method for the preparation of samples of human tissues with endometrial cancer by the use of the QuEChERS technique. Analytical parameters such as centrifugation temperature, extraction solvent, and adsorbents were modified to obtain satisfactory recovery for ZEA (R = 82.6%, RSD = 2.9%) and one of its metabolites, α-zearalenol (R = 50.1%, RSD = 3.2%). High-performance liquid chromatography (HPLC) with fluorescence detection and tandem mass spectrometry (LC-QTOF-MS) were used for the identification and quantitative determination of the analyzed compounds. The developed procedure was applied for analyses of human tissues with endometrial cancer. The presence of α-zearalenol was detected in 47 out of the 61 examined tissue samples.

Abstract:A comprehensive review is presented on the recent developments and trends in the QuEChERS (quick, easy, cheap, effective, rugged, and safe) sample preparation approach. This technique involves liquidliquid partitioning using acetonitrile and purifying the extract using dispersive solid-phase extraction (d-SPE). Originally, the QuEChERS was introduced for pesticides residues analysis in high moisture fruits and vegetables, but more recently it is gaining significant popularity in the analysis of broad spectrum of analytes in huge variety of samples. The wide range of the technique applications is possible due to introducing various modifications based on the use of different extraction solvent and salt formulation and buffer additions for salting-out partitioning step and the application of various d-SPE sorbents for clean-up step. Therefore, the QuEChERS approach is useful for analysis of, among others pesticides, veterinary drugs and other pharmaceuticals, mycotoxins, polycyclic aromatic hydrocarbons (PAHs), dyes, acrylamide, synthetic musks and UV filters, bisphenols, polybrominated diphenyl ethers and other flame retardants, endocrine disruptors, and other chemical compounds. Thanks to the QuEChERS approach, high-throughput multiresidue methods operate in a routine contaminant control of food products, feedstuff, and environmental samples.

The validation of a rapid, reliable and sensitive method for the simultaneous determination of Aflatoxins (AFB 1 , AFB 2 , AFG 1 , AFG 2 ), Ochratoxin A, Zearalenone, Deoxynivalenol, Fumonisins B 1 and B 2 , T-2 and HT-2 toxins, in nuts (peanuts, pistachio and almonds) and cereals (maize and wheat) is reported. The method was developed and validated to fulfil the requirements of the official control of mycotoxins according to EU legislation. This method is based on a single extraction step using an acetonitrile/ water mixture followed by the analysis of the diluted crude extract using ultra high performance liquid chromatography with tandem mass spectrometry (UPLC-MS/MS). The MS/MS detection was carried out using an electro spray-ionization interface (ESI) in positive ion mode. Matrix-matched calibration was used for the quantification of the mycotoxins, because of the absence of further clean up steps that reduce suppression/enhancement matrix effects. The method performance characteristics were determined after spiking blank samples on multiple levels. The mean recoveries of mycotoxins in spiked nuts ranged from 74.4% to 131.7%, while in cereals ranged from 52.8% to 113.9%. Relative standard deviations were lower than 20.4% for all target mycotoxins. Limits of detection and quantification for nuts and cereals ranged 0.08-30.0 and 0.25-99.0 μg/Κg, respectively.
