A new approach for treatment of precancerous lesions with curcumin solid–lipid nanoparticle-loaded gels: in vitro and clinical evaluation

Abstract: Objective: Preparation and characterization of curcumin solid-lipid nanoparticle (CurSLN)-loaded mucoadhesive gel for local treatment of oral precancerous lesions with low dose. Methodology: The formulated CurSLNs were dispersed in a mucoadhesive gel matrix to be applied to the buccal mucosa. Conventional mucoadhesive gel using binary system was adopted. The prepared gels were evaluated for in vitro drug dialysis, ex vivo mucoadhesion test and ex vivo permeation study using chicken buccal mucosa. Short-term cl… Show more

“…In vitro drug release profile of SA-NPs was investigated via the dialysis method (Hazzah et al, 2016), and DXM was determined using HPLC assay. Phosphate buffer (PBS, pH 7.4) was used as dissolution medium.1 mL DXM-loaded SA-NPs was sealed in a dialysis membrane (MWCO: 3500 Da), and then immersed into 40 mL release medium, free DXM and DXM-loaded NPs (equal DXM) as control.…”

“…57,58 SLNs have been extensively used in many studies to orally deliver bioactive compounds with an enhanced anticancer effect. 17,57,[59][60][61][62] Genistein is a phytoestrogen that is extensively used for hormone-related cancers, and it has limited bioavailability. Recently, genisteinloaded solid lipid microparticles (SLMs) with a particle size of 6 µm were compared with SLNs with a particle size of about 120 nm in terms of their bioavailability.…”
