A new anti-inflammatory β-carboline alkaloid from the hairy-root cultures of Eurycoma longifolia

Abstract: One new β-carboline alkaloid 7-methoxy-(9H-β-carbolin-1-il)-(E)-1-propenoic acid (1) together with 9-methoxycanthin-6-one (2) and 9-hydroxycanthin-6-one (3) were isolated from the hairy-root cultures of Eurycoma longifolia. The effects of these compounds on lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW264.7 cells were investigated. Compound 1 strongly inhibited the production of NO while 2 and 3 having weak or inactive effect. Consistently, compound 1 decreased the expression of cyclooxy… Show more

“…The results indicated the potential of a number of β-carboline-type alkaloids and canthinone-type alkaloids in the compound library as anti-inflammatory agents. Compounds ( 21 , 46 , and 57 ) have been reported with inhibitory NO production activities [2123], but compounds ( 11 – 13 , 15 , 22 , 23 , 27 , 29 – 31 , 37 , 41 , 42 , 47 , 55 , 64 , 71 , 72, and 76 ) were not reported as inhibitors of NO production.…”

“…7‐methoxy‐(9H‐β‐carbolin‐1‐il)‐( E )‐1‐propenoic acid (7‐MCPA) was isolated from the hairy root cultures of E. longifolia as previously described [Ngoc et al, ]. The chemical structure is shown in Figure .…”

“…The chemical structure is shown in Figure . 7‐MCPA was obtained as amorphous yellow powder, showed a [M + H] + peak at m/z 269, by electron spray ionization‐mass spectrometry [Ngoc et al, ] and the purity of 7‐MCPA was checked by 1 H and 13 C NMR spectra [Ngoc et al, ]. 7‐MCPA was solubilized with dimethyl sulfoxide and used at a final concentration of less than 0.05% dimethyl sulfoxide.…”
