A Nematode Larval Motility Inhibition Assay for Screening Plant Extracts and Natural Products

Abstract: An in vitro nematode larval motility inhibition assay has been developed to screen plant extracts for anthelmintic activity against the sheep parasite Trichostrongylus colubriformis. The usefulness of this assay was verified by results for extracts of the liverwort Plagiochila stephensoniana and the shrub Pseudowintera colorata. The activity of these extracts was due to 4-hydroxy-3‘-methoxybibenzyl (1) (IC50 0.13 mg/mL) and polygodial (2) (IC50 0.07 mg/mL), respectively. Synthetic analogues of 1 displayed enh… Show more

“…It has been shown to be effective against H. contours and T. colubriformis (Bennet-Jenkins and Bryant, 1996;Lorimer et al, 1996). These effects are attributed to the presence of tannins/ polyphenols in eucalyptus.…”

“…Two-fold dilutions (six) of each extract were prepared starting from 2000 µg/mL, which corresponds with the concentration range tested in previous studies by Lorimer et al (1996), Paolini et al (2004), Houzangbe-Adote et al (2005), and Eguale et al (2006). Each experiment was examined in triplicate, with the exception of larvae third stage positive controls, which were tested in duplicate.…”

“…The larval migration inhibition (LMI) bioassay was used in order to measure inhibiting activity against infective larvae as previously described by Rabel et al (1994), Lorimer et al (1996), and Houzangbe-Adote et al (2005). Briefly, the L3 larvae hatched in vitro from T. colubriformis eggs previously obtained from infected sheep were applied at a concentration of 30-40 for each well and incubated for 3 h at 20…”

“…Because of developing resistance to the existing anthelmintic drugs (Prichard 1990), there is a need for new anthelmintic agents. The anthelmintic activity of plant extracts can arise either from a direct action of a drug on the worms or through induction of gastrointestinal irritation and diarrhea (Lorimer et al 1996).…”
