A nanomedicine-promising approach to provide an appropriate colon-targeted drug delivery system for 5-fluorouracil

Abstract: Targeted drug delivery plays a significant role in disease treatment associated with the colon, affording therapeutic responses for a prolonged period of time with low side effects. Colorectal cancer is the third most common cancer in both men and women with an estimated 102,480 cases of colon cancer and 40,340 cases of rectal cancer in 2013 as reported by the American Cancer Society. In the present investigation, we developed an improved oral delivery system for existing anticancer drugs meant for colon cance… Show more

“…5-Fluorouracil is a chemotherapeutic drug used in the treatment of various cancers. The delivery of this drug causes serious side effects in the body, such as mucositis, alteration in the normal microflora environment of the colon, and diarrhoea [ 51 ]. For the target-specific treatment of colon cancer, 5-fluorouracil was encapsulated within XG-poly(-vinylpyrrolidone-co-poly) (PVP)—acrylic acid-based hydrogels.…”

“…Conventional chemotherapy has been effective to some degree, the key disadvantages of chemotherapy include inadequate bioavailability, large dosage criteria, harmful side effects, low therapeutic indexes, non-specific targeting and production of multiple drug resistance [93]. The positive effectiveness and negligible side effects of managed drug delivery vehicles have been preferred.…”

“…Given the systemic toxicity of many chemoand immunotherapeutic drugs, probiotic bacteria have been engineered to locally deliver the treatment at the tumor site (52)(53)(54) also by exploiting the affinity of anaerobic strains for the anaerobic tumor microenvironment (55). This has the double advantage of increasing drug efficacy and reducing adverse effects (52)(53)(54). In addition, probiotics strains have also been used to deliver anticancer proteins at the tumor site in order to inhibit its growth (55).…”
