The present study is aimed to investigate the extractability of bioactive compounds from mandarin (Citrus reticulata Blanco) peel using a pulsed mode of ultrasonic treatment. Box-Behnken experimental design associated with response surface methodology (RSM) was used for the process optimization. Ultrasound amplitude (60%-80%), time (5-15 min), and duty cycle (.30-.90) were the independent variables investigated to maximize the yield of total phenolic content (TPC), total saponin content (TSC), total flavonoid content (TFC), total condensed tannin (TCT), and antioxidant activity (DPPH and FRAP methods). The optimum predicted values of the TPC (27.23 g GAE/100 g dry weight [DW]), TFC (5.64 g QE/100 g DW), TCT (3.17 mg CE/g DW), TSC (42.36 mg EE/g DW), DPPH (4.01 mg GAE/g DW) and FRAP (13.71 mg TE/g DW) were obtained at conditions: 11.19 min of sonication at a duty cycle of .30 and 77.70% amplitude. Furthermore, in silico investigations of anti-oxidant properties of major flavonoids were done using molecular docking. The analysis showed a better binding affinity of narirutin for Cytochrome P450 and myeloperoxidase, while hesperidin showed a better binding affinity with lipoxygenase.The results obtained from the study showed better phenolic extraction and high antioxidant activity using pulsed mode sonication. The high binding scores of hesperidin and narirutin for all three key enzymes, as obtained by molecular docking, have shown their therapeutic potential in the development of antioxidant drugs. Practical ApplicationsA practical insight of the present research suggests the applicability of optimized pulsed mode extraction conditions for industrial processing of Citrus reticulata Blanco peel, a green technology useful in terms of its biomedical application. The optimized extraction conditions will give a higher yield with reduced time and with less use of extraction solvents. The in silico analysis provides scope for predictive modeling of novel antioxidative drug development using major flavonoid compounds of Citrus reticulata Blanco peel.

The present work aimed to analyze the drying kinetics and color degradation of moringa leaves in selected drying methods. Also, quantitatively analyze the retention of phytochemicals and antioxidant capacity. Moringa leaves were dried using three methods, namely, freeze‐drying (−42 ± 2°C, 0.024 ± 0.001 mbar), microwave drying (200, 400, and 600 W), and hot air oven drying (55, 65, and 75°C). Drying kinetics revealed that the drying of moringa leaves occurred in the falling rate period, which means the diffusion process controlled the moisture removal from leaves. The color degradation was most minor in freeze‐dried leaves, followed by microwave‐dried leaves at 200 W. The value of total phenolic content was found maximum (57.42 ± 2.49 mg GAE/g dm) under microwave drying at 200 W, while the same was found least in quantity (24.62 ± 1.10 mg GAE/g dm) for samples dried under hot air oven drying at 65°C. A similar pattern was observed for total flavonoid content. The antioxidant capacity of the samples dried in the freeze dryer was highest at 5.94 ± 0.25 mg GAE/g dm. This research revealed that microwave drying at 200 W is best regarding the overall quality attributes of moringa leaves.Practical ApplicationsDrying is one of the essential processes in the food industry, for example, drying is crucial for the production of herbal medicines, nutritional supplements and extracts, leaf‐based culinary ingredients, aroma and essential oils, natural dyes and pigments, etc. It is used to increase the shelf‐life of agricultural produce; it facilitates the transportation and storage of food products. This paper may help in the selection of the drying method and condition for leaves drying to have desired characteristics in the final product like color, bioactive compounds, or antioxidant properties; May help in the trade‐off among quality, time, and cost of the production. Moreover, it may be helpful in the designing of the multi‐mode dryer which may produce superior quality products with great energy efficiency in less time. Which further facilitates the manufacturing of nutritionally rich food products.

Probe Ultrasonic-Assisted Extraction of Andrographolide Rich Extract from Andrographis paniculata: Processing Parameters Optimization Using Response Surface Methodology

The conventional extraction method has low extraction efficiency. This study aims to optimize the ultrasonic-assisted extraction conditions of andrographolide from A. paniculata. A ½ inch ultrasonic probe sonicator was used to extract A. paniculata. The effects of extraction time (X1), duty cycle (X2) and amplitude (X3) on the yield of andrographolide (Y) were investigated by using response surface methodology. Experiments were designed according to 3-level 3-factor Box Behnken Design. The concentrations of andrographolide were quantified by high-performance liquid chromatography. Results of analysis of variance showed the data were well fitted with the 2nd-order polynomial equation with a R2 value of 0.9969 and p-value less than 0.005. Optimum extraction conditions were found to be at 5 min, 11 % duty cycle, and 66 A. A checkpoint experiment was carried out based on the optimum conditions, validated highest andrographolide was 3.50 ± 0.17 w/w%. Response surface methodology was successfully applied to optimize the extraction processing parameters for yielding the andrographolide-rich A. paniculata extract. This study could be relevant for future ultrasonic-assisted extraction scale-up of andrographolide.
