A multistrategy approach for improving the expression of E. coli phytase in Pichia pastoris

Abstract: Phytase is an additive in animal feed that degrades phytic acid in plant material, reducing feeding costs, and pollution from fecal phosphorus excretion. A multistrategy approach was adopted to improve the expression of E. coli phytase in Pichia pastoris. We determined that the most suitable signal peptide for phytase secretion was an α-factor secretion signal with an initial enzyme activity of 153.51 U/mL. Increasing the copy number of this gene to four increased phytase enzyme activity by 234.35%. PDI overex… Show more

“…The present study obtained a final phytase activity of 12.65 MU/L which represented a 36-fold improvement compared to flask fermentations (Navone et al, 2021a). In instrumented fermenters, the reported commercial phytase production comprises levels from 5 MU/L to 30 MU/L (Helian et al, 2020;Luo et al, 2007;Mittal et al, 2013;Yao et al, 1998), suggesting the capability of our P.…”

“…The present study obtained a final phytase activity of 12.65 MU/L which represented a 36‐fold improvement compared to flask fermentations (Navone et al, 2021a). In instrumented fermenters, the reported commercial phytase production comprises levels from 5 MU/L to 30 MU/L (Helian et al, 2020; Luo et al, 2007; Mittal et al, 2013; Yao et al, 1998), suggesting the capability of our P. pastoris P1 strain to produce commercially relevant levels of phytase and the potential to continue improving phytase production by metabolic engineering optimization such as the improvement of the protein secretion capability of the α‐mating factor secretion signal (Helian et al, 2020). The specific activity of E. coli phytase has been reported to be around 3.6 MU/g (of pure phytase) (Li et al, 2015), which was similar to the specific phytase activity of 4 MU/g obtained in this study after the de‐repression phase (Figure 2 and Table 3).…”

“…After that, the ER-associated degradation pathway can be activated and then reduce the production of heterologous proteins. Based on the above rationales, to prevent the degradation of overexpressed heterologous protein by the proteasome, the disruption of Pep4 protease increases phytase secretion in P. pastoris [ 12 ]. Lipase is widely used in the production of biodiesels [ 13 ], detergents [ 14 ], chiral compounds [ 15 ], etc.…”

“…One unit of pectinase activity is the amount of enzyme that releases 1 μmol of d ‐(+)‐galacturonic acid per min in the buffer (pH 8.0) at 40 °C. The activities of tannase and phytase were determined by Ma’s (Ma et al ., 2015) and Helian’s (Helian et al ., 2020) methods.…”
