A Multiplex Immunoassay for Human Adipokine Profiling

Abstract: BACKGROUND:Adipose tissue secretory proteins, called adipokines, play pivotal roles in the pathophysiology of obesity and its associated disorders such as metabolic syndrome, type 2 diabetes, and cardiovascular disease. Because methods for comprehensive adipokine profiling in patient plasma and other biological samples are currently limited, we developed a multiplex immunoassay for rapid and high-throughput measurement of 25 adipokines in only 50 L of sample.

“…For a broader investigation of biomarkers, some working groups applied multiplex cytokine measurements in the blood samples of MetS patients and reported a range of cytokine concentrations comparable to those measured in our study (Schipper et al, 2010;Bremer et al, 2011;Sharabiani et al, 2011;Surendar et al, 2011;Tisato et al, 2013). We identified IL-6 and IL-10 to be significantly elevated in MetS patients.…”

“…These data highlight a specific low-grade inflammation in obesity/MetS, with the majority of cytokine levels tested not being increased in blood. However, the reports show some variation related to gender (Saltevo et al, 2008), ethnicity (Morimoto et al, 2014), sample size (Schipper et al, 2010;Phillips & Perry, 2013), clinical heterogeneity of MetS (Phillips & Perry, 2013), and different definitions of MetS (Surendar et al, 2011;Tisato et al, 2013). Specifically, increased waist circumference is mandatory in the IDF definition, while being just one facultative parameter in the NCEP definition (Expert Panel on Detection, Evaluation, and Treatment of High Blood Cholesterol in Adults, 2001;Alberti et al, 2009).…”
