A Multifunctional Platform for Tumor Angiogenesis-Targeted Chemo-Thermal Therapy Using Polydopamine-Coated Gold Nanorods

Abstract: Image-guided combined chemo-thermal therapy assists in optimizing treatment time, enhancing therapeutic efficiency, and circumventing side effects. In the present study, we developed a chemo-photothermal theranostic platform based on polydopamine (PDA)-coated gold nanorods (GNRs). The PDA coating was thin; however, it significantly suppressed the cytotoxicity of the cetyltrimethylammonium bromide template and allowed high cisplatin loading efficiency, arginine-glycine-aspartic acid (RGD) peptide (c(RGDyC)) con… Show more

“…First, the Bi 2 S 3 @mPS NPs reacted with (3-aminopropyl) triethoxysilane (APTES) for 48 h in ethanol to form NPs terminated with an amino group. Then the resultant Bi 2 S 3 @mPS-NH 2 reacted with NHS-PEG-Mal via Michael addition reaction [25] to form Bi 2 S 3 @mPS-PEG-Mal NPs. The activated Bi 2 S 3 @mPS-PEG-Mal NPs were then covalently coupled to thiolated Tam (SH-Tam) through thiol-maleimide linkages.…”

“…Obviously, particle size and shape effect along with particle surface charge/modification effect should be also considered together, and all these affect biodistribution, targeting efficiency, circulation time, and excretion in a collective manner. [120] Furthermore, PTI can be combined with photothermal therapy (PTT) under the same light radiation; thus, it can offer not only easy accessibility for multimodal molecular imaging but also an efficient tool for real-time imaging-guided therapy. [69] Up to now, various types of plasmonic nanoparticles have been applied to PTI, and PTI has garnered much attention in biomolecular imaging due to its potential in sensitivity, realtime monitoring, rapid recording with simple instruments and convenient use.…”

“…Moreover, it is not affected by photobleaching. All these features make lifetime‐resolved imaging an accurate and reliable method to measure the microenvironmental changes of a fluorophore 9. Owing to their high spin–orbit coupling associated with the heavy metal ion, metal complexes are ideal candidates for phosphorescence lifetime imaging microscopy (PLIM) with emission lifetimes up to microsecond that can totally deplete the background emission (usually below 10 ns).…”
