A mouse homeo box - containing gene maps near a developmental mutation

Abstract: Mouse genes containing homeo box domains are predicted to fulfill important functions in embryogenesis. Using recombinant inbred mouse strains, we have mapped a mouse gene which contains a homeo box with homology to the Drosophila engrailed gene. This gene maps to mouse chromosome 1 near or at the dominant hemimelia locus which is a known mouse developmental mutation.

“…match cleavage technique was used to look for any base Analysis of Gli2 Coding Sequence changes (Condie et al, 1993). An anomalous fragment In the absence of gross genomic abnormalities and was observed in fragment 1, and sequence analysis of PCR products identified a silent base change in both changes in expression levels of Gli2 in Dh, we analyzed et al, 1994), and c (Hill et al, 1987).…”

“…In the mouse, this complex is located on chromosome 2 [27]. However, the mouse En-2 gene is located on chromosome 1 [28], and the long arm of human chromosome 2 corresponds to mouse chromosomes 1 or 2, as indicated by comparative genome mapping [18,19]. In fact, human EN-1 is located on chromosome 2 [21].…”

“…No crossovers were detected between Tgfb-2 and Hlx, giving 95% confidence that they lie within 1.6 cM (probably within a few megabases). The placement of At-3, Spna-1, and Tgfb-2 relative to other chromosome 1 markers on the interspecific and composite intraspecific backcross maps (see Dickinson et al 1990) places Hlx well below the loci for the En-1 (Hill et al 1987;Joyner and Martin 1987) and Oct-1 (Hsieh et al 1990) homeo box loci. Interspecific backcross analysis on the same panel maps Copeland, and N.A.…”
