A Monoclonal Antibody Against Transmissible Gastroenteritis Virus Generated via Immunization of a DNA Plasmid Bearing TGEV S1 Gene

Abstract: Transmissible gastroenteritis virus (TGEV) is a member of the coronaviruses. The viral spike (S) protein of TGEV mediates interaction between TGEV and its susceptible cells. Herein, DNA plasmid bearing TGEV S1 gene (the N terminal half of TGEV S gene) was used to immunize BALB/c mice followed by generation of a monoclonal antibody (MAb) using the hybridoma technique. The generated MAb (1H4) was identified by ELISA. Immunofluorescence assays showed that MAb 1H4 was able to detect infection of cells with TGEV. T… Show more

“…Coronaviruses (CoVs) of late have been very common throughout the world and among the different types of CoVs, Severe Acute Respiratory Syndrome Coronavirus (SARS-CoV) andthe Middle East Respiratory Syndrome Coronavirus (MERS-CoV) have been found to be most deadly of them all. A particular form of porcine coronavirus known as Transmissible gastroenteritis virus (TGEV) 68,69 had caused increased mortality in pigs and the vaccination to piglets provided protection to suckling piglets, however, on a larger scale, it did not promote enhancement of immunity and did not control the spread of the infection. 70,71 Hence the development of an alternative therapeutic strategy is imminent in the control of such an infection.…”

“…Particularly some coronaviruses, such as severe acute respiratory syndrome coronavirus (SARS-CoV) and the recently identified Middle East respiratory syndrome coronavirus (MERS-CoV) may cause fatal infection in human. Transmissible gastroenteritis virus (TGEV), a porcine coronavirus, causes very high mortality in piglets and thus has tremendous impact of pig industry [11,12]. Vaccination has been extensively applied to prevent pigs from TGEV infection.…”
