A monoclonal anti-DC1 antibody selectivity inhibits the generation of effector T cells mediating specific cytolytic activity.

Abstract: The products of the DC locus have been shown to be structurally different from those of the DR locus. In this paper it is shown that, unlike anti-DR antibodies, a monoclonal antibody directed against DC1 does not affect proliferation of T cells in response to alloantigens or soluble antigens or production of Ig in a pokeweed mitogen-stimulated in vitro culture. However, the anti-DC1 inhibits the generation of effector T cells mediating specific cytolytic activity, whereas no inhibitory effect can be observed o… Show more

“…In this respect, it has been found that only the DR+, DC+ and not DR+, DC-subpopulation of human monocytes is able to function as APC for autologous T lymphocytes (87) and that DC molecules can play a role in the generation of cytotoxic lymphocytes in an MLC (88). Following these observations, it was of interest to determine first the relative expression of DR and DC products on Me cells, and then the possibility to modulate them by IFN-y.…”

“…Further dissociation of the functional properties of HLA-DR and HLA-DQ antigens have been demonstrated. Corte et al (1982) showed that an anti-DQwl Moab was capable of blocking the production of specific cytotoxic cells in MLC. No blocking was obtained with a DR Moab.…”

“…The results of Sasazuki et al (1984) on suppressor T cell dependent low responsiveness to Streptococcal antigens associated with DQwl and data from Corte et al (1982) on the inhibition of cytotoxic T cells with the help of a monoclonal anti-DC1 (DQwl) antibody support the latter (suppressorkytotoxic) hypothesis. If DQwl is involved in the DRw6 donor effect, we would expect the same phenomenon with the D R 1 and DR2 positive grafts which are also in most cases DQwl positive.…”
