A Modified Form of Diphthamide Causes Immunotoxin Resistance in a Lymphoma Cell Line with a Deletion of the WDR85 Gene

Abstract: Background: Some children with leukemia do not respond to an immunotoxin containing Pseudomonas exotoxin A. Results:We isolated a resistant cell line and showed that a WDR85 gene deletion causes immunotoxin resistance by allowing formation of diphthamide containing an extra methyl group. Conclusion: Low WDR85 expression can cause immunotoxin resistance in patients. Significance: Analysis of EF2 may reveal why some patients fail therapy.
