A Model of Isolated Autologously Hemoperfused Porcine Slaughterhouse Kidneys

Abstract: Background: The rapidly evolving field of transplantation research with a focus on ischemic and reperfusion injuries has gained importance since the methodology of organ preservation significantly limits graft survival. Numerous models of isolated perfused kidneys have been established in the past years but limitations such as organ size, perfusate and ethical standards have restricted a widespread research in this area. Methods: A model of hemoperfused isolated porcine slaughterhouse kidneys was established w… Show more

“…Isolated kidney perfusion was performed in an ex vivo model with normothermic autologous blood [8, 9, 10]. Kidneys were obtained either from anesthetized pigs after clamping the renal vessels in the laboratory or from pigs sacrificed at the slaughterhouse.…”

“…5 -7,14 In order to reduce the number of animal experiments, the use of NHBD from a commercial abattoir has been proposed. 5,7 The exploitation of slaughterhouse organs would be a crucial step forward in reducing the number of animal experiments since the use of slaughterhouse kidneys is not legally defined as an animal experiment and would fulfil international standards as an alternative to animal experiments. 18 The subjection to warm or cold ischaemia-and reperfusion-related injuries impairing the functional and structural integrity of donor organs is still a general problem.…”

“…Before sterile transfer of the organs on ice, preservation was established by infusion of 500 ml preservation solution (table 1). After cold storage (t-kons; table 3), 30 kidneys (15 B2 and 15 HTK) were perfused as described [18]. Tissue biopsies were taken from the other 30 kidneys after 2, 8 and 24 h of cold storage for the characterization of early histopathological changes during cold storage (fig.…”

“…Inside the dialysis circuit which contains 6,000 ml of dialysate medium [18], a roller pump (duo-head Stöckert 10-20-00) pumps the dialysate from the dialysate reservoir to the dialysis model (1,000 ml/m). The perfusate-dialysate relation is regulated on the basis of the amount of produced urine.…”
