A Model for the Low-Spin, Non-Heme, Thiolate-Ligated Iron Site of Nitrile Hydratase

Abstract: Nitrile hydratase is the first and only current example of a metalloenzyme containing a single non-heme Fe in a mixed N/Sligated coordination site.'-3 Its function is to hydrolyze nitriles to the corresponding amides in organisms which can live on R-CN as their sole C and N source. On the basis of EPR? MCD,5 resonance Raman, EXAFS,* and ENDOR' studies, the active site of nitrile hydratase is proposed to contain a six-coordinate, low-spin (S = ' / 2 ) Fe3+ ion ligated by two ciscysteinates, three N's, and a wat… Show more

“…45,56 Like the NHase active site, 7,11 complex 3 is low-spin (S = ½) over a wide temperature range, stabilized in the 3+ oxidation state (E 1/2 = −1.01 V vs SCE), and intense green in color (λ max = 718 (2500) nm). A low spin state is unexpected for non-heme iron, especially when ligated by π-donors such as RS − , which tend to decrease 10Dq.…”

“…The structure of monocationic 3 was described previously. 23 Metrical parameters for 1-4 are summarized in Table 2. 29 The iron in structures 1-4 is contained in a pseudo-octahedral bis(tris-chelated) environment, ligated by two trans-imines, two cisthiolates, and either terminal pyridines (py) (1), imidazoles (Im) (2), or amines (NH 2 ) (3 and 4).…”

“…The structure of the iron center in the active form has been studied by various spectroscopies including ESR (3), resonance Raman (13), extended x-ray absorption fine structure (13) and electron nuclear double resonance (14), and the ligand-donor set of N 3 OS 2 has been proposed (14), which is supported by model complexes of the iron center (15)(16)(17). Recently, the metal site structure has been studied in detail by means of electron nuclear double resonance (18), resonance Raman (19), and x-ray absorption (20) spectroscopies.…”
