A Mitochondrion‐Specific Photoactivatable Fluorescence Turn‐On AIE‐Based Bioprobe for Localization Super‐Resolution Microscope

Abstract: A novel mitochondrion-specific photo-activatable fluorescence turn-on bioprobe, named as o-TPE-ON+, is designed and readily prepared, operating through a new photoactivatable mechanism of photocyclodehydrogenation. This bioprobe exhibits unique photoactivation behavior in cells, and is applied to super-resolution imaging of mitochondrion and its dynamic investigation in both fixed and live cells under physiological conditions without any external additives.

“…The double bonds in these AIEgens may also lead to potential photobleaching or photooxidation which will result in a fluorescence loss. 20,21 As well as these drawbacks in AIEgens with double bonds, some shortcomings also exist in other propeller-like AIEgens. For example, silole and its derivatives, HPS and DMTPS, are usually hard to prepare and are unstable under alkaline conditions.…”

“…Subsequently, another fluorescence turn-on photoactivatable AIE mitoprobe of o-TPE-ON + with yellow emission was reported, [73] which could be photoactivated after transforming into c-TPE-ON + through the same mechanism as previous one ( Figure 4A). The probe could efficiently light up the mitochondria in living and fixed HeLa cells upon UV light irradiation.…”

“…To solve the problem with ACQ effects, a group of AIEgens has been successfully designed for mitochondria targeting and imaging . An AIE‐based dye, TPE‐Ph‐In, which is a positively charged cationic molecule, could be simply synthesized by incorporating indolium salt into the TPE unit ( Figure ).…”
