A Method to Assess Genomic DNA Methylation Using High-Performance Liquid Chromatography/Electrospray Ionization Mass Spectrometry

Abstract: Eukaryotic DNA is methylated at some cytosine residues, and this epigenetic feature performs critical functions. We developed a method for quantitative determination of 5-methyl-2'-deoxycytidine in human DNA using liquid chromatography/electrospray ionization mass spectrometry (LC/ESI-MS). The DNA was enzymatically hydrolyzed by sequential digestion with three enzymes. DNA hydrolyzates were subsequently separated by reversed-phase high-performance liquid chromatography in isocratic mode. The four major DNA bas… Show more

“…Another intriguing example of disease susceptibility related to methyl donor diet was illustrated in a population study reporting an association between folate status and a mutation in the gene encoding methylenetetrahydrofolate reductase, a key enzyme in determining genomic DNA methylation. This study suggests an interplay between nutritional epigenetics and genetic susceptibility in the modulation of gene expression [51]. Collectively, these findings offer valuable insights into the pathophysiology of diseases associated with nutritional epigenetics.…”

“…For HPLC separation of nucleosides, cells were split 1:3 into tracer medium in 100-cm 2 culture plates, grown to confluence, and harvested. DNA was digested to nucleosides prior to separation by HPLC, using procedures described elsewhere (28). Peaks corresponding to nucleosides were identified from standards, collected, and isotope levels quantified.…”
