A method for the direct isolation of IncH plasmid-dependent bacteriophages

Abstract: A method is described for the isolation of IncH plasmid‐dependent bacteriophages present in very low numbers in sewage. An isogenic plasmid‐lacking host was used to absorb the majority of non‐specific phages. This resulted in an enrichment of 100‐to 400‐fold of the IncH‐dependent phages. A total of 27 IncH‐dependent phages were isolated and all were shown to be temperature‐sensitive RNA phages which produced turbid plaques.

“…The recipient strain was E. coli K-12 strain J53-1 (pro met F-Nal) (2). Bacteriophage Hgal (21) is an H-pilus-specific RNA phage, 18.7 nm in size (16), which adsorbs to the sides of the H-pilus shaft.…”

“…The elevated pilus levels associated with the R27::TnlacZ derivative make it a suitable test plasmid to study H-pilus assembly kinetics at permissive and nonpermissive conjugation temperatures. Two single-stranded RNA bacteriophage, pilHa and Hgal (10,16,21), adsorb uniformly along the H-pilus shaft. The goal of this study was to exploit phage adsorption, constitutive pilus production, and the thermosensitive conjugation system of R27::TnlacZ to examine several aspects of H-pilus assembly, stability, and morphology by transmission electron microscopy (TEM) and field emission scanning electron microscopy (FESEM).…”

“…Phage C-1 was originally isolated from sewage samples in Pretoria, South Africa (55), and phage Hgal1 was also isolated from raw sewage samples collected from the sea outfall in Galway docks, Ireland (43). Since their initial isolation and characterization, no further work has been performed.…”

“…Phage PRR1 [16] which adsorbs specifically to IncP plasmid-encoded pili was the first such example, and later other phages specific for Inc group C [17], D [18], H [19,20], I [21], M [22] and T [23] plasmids followed. Phages PRR1, C-1 (IncC-specific) and Hgal1 (IncH-specific) have been sequenced [24,25] and phage PRR1 capsids have also been crystallized [26], but no research has been done on the other plasmid-specific phages since their isolation.…”
