A Method for Enzymic Extraction and the Measurement of Chloroplast RNA

Abstract: Smillie and Krotkov (23) compared various methods for quantitating RNA in plant materials, and concluded that the SchmidtThannhauser procedure (20), which involves repeated tissue extractions with acid and organic solvents followed by KOH hydrolysis and use of an anion exchange resin, is the most reliable method for the widest range of plant tissues. De Deken-Grenson and de Deken (9) made the same conclusion, and described a batch method for the use of Dowex 2 in purifying the free nucleotides resulting from K… Show more

“…For this procedure to be valid, however, the total RNA has to consist very largely of the rRNA of 70S ribosomes. Nonribosomal RNAs were found to be an insignificant fraction of the total RNA bound to thylakoids (10). Contamination by 80S ribosomes is essentially absent from intact chloroplasts prepared on Percoll gradients (24) and from the thylakoids extracted from them (Fig.…”

“…We used the convenient and rapid enzymic method for measuring total RNA (10) to estimate the number of ribosomes bound to thylakoids, because there is no simple, quantitative method to obtain this estimate more directly. For this procedure to be valid, however, the total RNA has to consist very largely of the rRNA of 70S ribosomes.…”
