The influence of essential oils (EOs) from medicinal and aromatic plants from sage (SA), cinnamon (CN), thyme (TH) and oregano (OR) on the amylolytic, proteolytic and cellulolytic activities in chyme of the duodenum (DU), the small (SI) and large intestine (LI), the caecum (CE) and the rectum (RE) as well as on the growth ability of laboratory ICR outbred mice were compared in four feeding trials. The negative control was present in the each trial. EOs were mixed into a feed mixture (crude protein (CP) 170.0, fibre 115.0, fat 27.0, lysine 7.0, methionine and cystine 6.7, Ca 9.0, P 6.0 g.kg-1 dry matter (DM), metabolic energy (ME) 10 MJ.kg-1 DM) of experimental group as follows: 1) 6 groups (n=36, age 63 days, period 14 days) SA, CN, TH, OR, the blend of SA with OR, the dosages of EOs 0.42 except OR 0.21 ml.100 g-1 feed, 2) 2 groups (n=12, age 28 days, period 30 days) blend of SA 0.42 with OR 0.21 ml.100 g-1 feed, 3) 3 groups (n=18, age 28 days, period 58 days) CN and TH, both 0.5 ml.100 g-1 feed, 4) 2 groups (n=12, age 28 days, period 8 days) the blend of CN with TH 0.42 ml.100 g-1 feed. The peroral intake of blend of EOs from OR with SA increased the weight gains by 25 %. Additionally, it stimulated the activities of digestive enzymes in the chyme of intestinal apparatus of laboratory mice in the experimental group compared to control as follows: amylolytic by 4,138 µmol.s-1.g-1 and proteolytic by 282.2 mg azoalbumin.min-1.g-1 in SI (p<0.01), cellulolytic by 23.58 in LI and by 34.87 mmol glucose.min-1.g-1 in CE (p<0.01).

Extraction and isolation of polyhydroxy triterpenoids from Rosa laevigata Michx. fruit with anti-acetylcholinesterase and neuroprotection properties

Rosa laevigata fruit, at present, is becoming increasingly popular as a functional foodstuff with several nutritional and medicinal properties. To explore the acetylcholinesterase (AChE) inhibitory activity of extracts from the Rosa laevigata Michx. fruit (RLMF), a simple and efficient enrichment purification technology based on microwave-assisted extraction (MAE) and multi vacuum extraction columns (VEC) was applied to screen and identify triterpenoids (TTs) in the RLMF extracts. The MAE conditions were optimized using the Box-Behnken design (BBD) with a quadratic regression model and the response surface method (RSM). The optimum conditions were as follows: ethanol concentration, 69%; extraction time, 12 min; ratio of liquid to raw material, 26 : 1 mL g À1 ; and microwave power, 528 W. Under these conditions, the maximum content of triterpenoids reached 62.48 AE 0.25 mg g À1 , which was close to the predicted value of 62.69 mg g À1 . In addition, two pure polyhydroxy triterpenoids: 2a,3b,19a,23tetrahydroxyurs-12-en-28-oic acid (1) and 2a,3b,19a,23-tetrahydroxyurs-12-en-28-oic acid-28-O-b-Dglucopyrannoside (2) were isolated and enriched to more than 500 mg by a multi VEC method.Furthermore, the quantities of compounds 1 and 2 from RLMF were 5.36 and 10.37 mg g À1 , respectively, as determined using HPLC. These compounds were further assessed for acetylcholinesterase inhibitory and neuroprotection properties. The results showed that 1 and 2 showed potent AChE inhibitory activities with IC 50 values of 29.22 and 45.47 mg mL À1 , respectively. At high concentration, compounds 1 and 2 produced a 92% and 89% inhibition on the target enzyme, which was consistent with docking results between AChE and each isolate. The chemical composition as well as antioxidant, antiproliferative, and enzyme inhibition activities of extracts from aerial parts of Thymus leucostomus H ausskn. & V elen. obtained with hexane, methanol, and water were evaluated. Results showed that the methanol extract had significantly (p < 0.05) the highest total phenolic content (TPC; 107.80 mg GAE/g) and total flavonoids content (TFC; 25.21 mg RE/g) followed by the aqueous extract (102.72 mg GAE/g and 20.88 mg RE/g, respectively). LC‐MS/MS‐guided profiling of the three extracts revealed that rosmarinic acid (34.8%), hesperetin (42.9%), and linoleic acid (18%) were the dominant compounds in the methanol, aqueous and hexane extracts, respectively. GC‐MS analysis of the hexane extract showed that ɣ‐sitosterol (29.9%) was the major constituent. The methanol extract displayed significantly (p < 0.05) the highest Cu++, Fe+++, and Mo(VI) ions scavenging and reducing properties while the aqueous extract exerted significantly (p < 0.05) the highest metal chelating power (42.51 mg EDTAE/g). Both the hexane and methanol extracts effectively inhibited the acetylcholinesterase enzyme (2.63 and 2.65 mg GALAE/g, respectively) while the former extract exerted significantly (p < 0.05) the highest butyrylcholinesterase (2.32 mg GALAE/g), tyrosinase (19.73 mg KAE/g), and amylase (1.16 mmol ACAE/g) inhibition capacity. The aqueous extract exhibited the best glucosidase inhibition property (0.49 mmol ACAE/g). The methanol and hexane extracts exerted a higher cytotoxic effect on HT‐29 (IC50: 8.12 µg/mL) and HeLa (IC50 = 8.08 µg/mL) cells, respectively. In conclusion, these results provide valuable insight into the potential use of T. leucostomus bioactive extracts in different pharmaceutical applications.
