A meta-analysis of data associating DRD4 gene polymorphisms with schizophrenia

Abstract: To explore the association between DRD4 polymorphisms and schizophrenia risk, a meta-analysis was carried out with 41 case–control articles. Specifically, we included 28 articles (5,735 cases and 5,278 controls) that pertained to the 48 bp variable number tandem repeat (VNTR) polymorphism, nine articles (1,517 cases and 1,746 controls) that corresponded to the 12 bp tandem repeat (TR), six articles (1,912 cases and 1,836 controls) that addressed the 120 bp TR, 10 articles (2,927 cases and 2,938 controls) that … Show more

“…16 We selected the most appropriate genetic model using a previous study. 21,22 We evaluated the degree of inter-study heterogeneity using a Q statistic, 23,24 where P > 0.05 was defined as an absence of heterogeneity. We performed subgroup analysis for ethnicity (e.g., Caucasian or Asian), source of controls (e.g., hospitalor population-based), and type of cancer (i.e., breast cancer, prostate cancer).…”

“…To assess the impact of each study on the pooled results, sensitivity analysis was performed by removing single studies in turn. The power was calculated using the PS program [26]. The threshold for statistical significance was P < 0.05 in all tests.…”

“…As described in a previous study, 26 OR1 (AA vs. aa), OR2 (Aa vs. aa), and OR3 (AA vs. Aa) were compared, where A is the risk allele, from which the most appropriate genetic model was selected. 27,28 A Q statistic was used to evaluate the degree of inter-study heterogeneity, with the absence of heterogeneity being defined as P > 0.05. 29,30 The I 2 is the proportion of observed variance in effect size attributable to the true differences among studies.…”
