A MALDI-TOF MS library for rapid identification of human commensal gut bacteria from the class Clostridia

Abstract: IntroductionMicrobial isolates from culture can be identified using 16S or whole-genome sequencing which generates substantial costs and requires time and expertise. Protein fingerprinting via Matrix-assisted Laser Desorption Ionization–time of flight mass spectrometry (MALDI-TOF MS) is widely used for rapid bacterial identification in routine diagnostics but shows a poor performance and resolution on commensal bacteria due to currently limited database entries. The aim of this study was to develop a MALDI-TOF… Show more

“…To maintain the extensibility of the application, the HDF5 file panel was created by storing the averaged spectra of each dataset, their annotation, the parameters used and their intensity matrix. This module will be helpful for the future construction of a reference MS spectra database which will be used for specimen identification by spectral matching [43].…”

“…The species were inoculated from frozen stocks into liquid culture media and passaged twice (1:100) overnight to ensure robust growth. The purity and identity of the species was regularly verified via sequencing of the 16S rRNA gene and/or MALDI TOF mass spectrometry (MS) 45 .…”

“…Each colony on the plate is treated with 1μl of 25% formic acid and subsequently with 1μl of α-Cyano-4-hydroxycinnamic (CHAC) matrix. After sample preparation, the plate is loaded into a Microflex MALDI-TOF instrument for spectra measurement (Bruker Daltonics) and subsequent identification [43].…”
