A Long Term Double-Blind Clinical Trial of Ibuprofen and Indomethacin in Rheumatoid Arthritis

Abstract: Two-hundred and eighteen individuals with rheumatoid arthritis were randomly assigned to six months treatment with ibuprofen (900-1800 mg/day) or indomethacin (75-150 mg/day). The drugs were equally effective in the treatment of rheumatoid arthritis while the incidence of indomethacin side-effects was 1.5 times greater than the incidence of ibuprofen side-effects.

“…Ibuprofen is a 2-phenylalkanoic acid derivative which is among the most commonly prescribed NSAIDs, indicated for analgesia, gout, osteoarthritis, and other rheumatic diseases (Flower et al, 1985;Kantor, 1979;Royer et al, 1975;Schweitz et al, 1978). Based on the data of previous published reports on the instability of the arylacetic acids to the light (Givens and Levi, 1979;Meiggs et al, 1972;Epling and Lopes, 1977), Ibuprofen, by reasons of chemical analogy, should be photolabile.…”

“…Underdosing was a problem in many of the early evaluations of ibuprofen where daily doses of 1,600 mg or less were used (12). We believe, however, that 2,400 mg daily should have been adequate to produce antiinflammatory effects, as have been demonstrated at that dose with rheumatoid arthritis (13), osteoarthritis (14), and gout (15). Not only was ibuprofen demonstrated to be less effective than aspirin, but it was found to be as toxic.…”
