A long-range physical map of human Chromosome 21q22.1 band from the YAC continuum

Abstract: The human Chromosome (Chr) 21q22.1 region contains several genes for cytokines and neurotransmitters and the gene for superoxide dismutase (mutant forms of which can cause familial amyotrophic lateral sclerosis). A region of approximately 5.8 Mb encompassing D21S82 and the glycinamide ribonucleotide transformylase (GART) loci was covered by overlapping YAC clones, which were contiguously ordered by clone walking with sequence-tagged site (STSs). A total of 76 markers, including 29 YAC end-specific STSs, were u… Show more

“…A cosmid contig of a 4-Mb region in the so-called DS critical region (see below) has been constructed (Osoegawa et al, 1996). Additional bacterial clone contigs for different regions of 21q have been constructed (e.g., Soeda et al, 1995;Lafrenière et al, 1995, Patil et al, 1995, Eki et al, 1996, Ohira et al, 1996Hubert et al, 1997). All of these maps of overlapping clones are necessary for the identification and characterization of HC21 genes that are involved in various disease phenotypes including those of DS.…”
