A kinetic study of sugarcane sucrose synthase

Abstract: The kinetic data on sugarcane (Saccharum spp. hybrids) sucrose synthase (SuSy, UDP-glucose: D-fructose 2-a-Dglucosyltransferase, EC 2.4.1.13) are limited. We characterized kinetically a SuSy activity partially purified from sugarcane variety N19 leaf roll tissue. Primary plot analysis and product inhibition studies showed that a compulsory order ternary complex mechanism is followed, with UDP binding first and UDP-glucose dissociating last from the enzyme. Product inhibition studies showed that UDP-glucose is … Show more

“…However, SPS activity becomes gradually more important in sucrose synthesis as the internodes get older, exceeding sucrose synthase more than threefold (Botha and Black 2000). The data from Schafer et al (2004) are in accordance with the labeling data from Botha and Black (2000) which clearly demonstrated the sucrose synthase involvement in sucrose synthesis in young internodes.…”

“…The sugarcane isozyme homologous to maize SS2 appeared to be down regulated in the older internodes, the one having highest level of endogenous sucrose. Schafer et al (2004) partially purified sucrose synthase activity from sugarcane leaf tissue. The product inhibition studies revealed that UDP-glucose is a competitive inhibitor with respect to UDP and a mixed inhibitor with respect to sucrose.…”

“…Although several cyanobacterial (8,16,19) and plant (14,17,(26)(27)(28)(29) sucrose synthases have been characterized, the enzyme from nonphotosynthetic bacteria has never been studied, and no structural information on any sucrose synthase from bacterial sources is available. In this work we report the recombinant expression and biochemical characterization of N. europaea sucrose synthase and its crystal structure.…”

“…Thus, while the increase in sucrose accumulation was correctly predicted by the model, its scope did not extend far enough to predict the effect of changes in neutral invertase activity on respiration and growth. This model has also been used by Schäfer et al (2004) to investigate the kinetics of one of the sucrose synthase isozymes, SuSy C. Bosch (2005) used the model to describe carbon partitioning to trehalose to determine if this affects sucrose accumulation; the study concluded that it does not. Wu & Birch (2007) showed that introducing an enzyme to convert sucrose to isomaltulose doubles the total sugar concentration, but does not affect the sucrose concentration.…”
