A kinetic method for assay of plant peroxidase and catalase activities by chemiluminescence

Abstract: A chemiluminescent method for the simultaneous assay of peroxidase and catalase activities and chemical traps and quenchers of free radicals in natural samples, based on the additions of peroxidase standards, is presented. The method was standardized with horseradish peroxidase and beef liver catalase. The theoretical basis, mathematical treatment and computer processing of the method are described. The technique was used to analyse 25 samples of various common fruits and vegetables. The levels of peroxidase a… Show more

“…Once the samples had been powdered, the powder was added to 1 mL of 50 mM sodium-phosphate buffer (pH 7) containing 2 mM dithiothreitol, 2 mM EDTA, 0.2% Triton X-100, and 50 mM Tris-HCl. The resultant sample was centrifuged at 12,000 rpm for 30 min at 4 • C, and the supernatant was used for measurement of CAT and APX activities [38]. Soluble protein was determined using bovine serum albumin (BSA) at 595 nm by a UV/VIS spectrophotometer according to the method described by Bradford [39].…”

“…5,6) This enzyme has also been used to study the role of reactive oxygen species (ROS) involved in gene expression and apoptosis. 7,8) Although catalase was one of the first enzymes to be isolated and purified, its physiological function and regulation are not yet fully understood.…”
